چکیده

background brucellosis is a zoonotic disease that is endemic in iran. appropriate and rapid diagnosis has a vital role in public health improvement. low isolation rate of the organism has reported frequently in various reports. the present study was conducted to determine the isolation rate of organism in culture from collected specimens of hospitalized patients who were not under antibiotic therapy. meanwhile, comparing the direct inoculation to biphasic media with lysis method was also determined. materials and methods twenty-five hospitalized brucellosis patients diagnosed on the basis of clinical manifestations and positive serologic tests were included. blood samples were provided and cultured either as direct inoculation into biphasic media or lysis method by washing with distilled water before culture on solid media. results brucella was isolated in 4 samples (16%). further studies revealed all these four cases to be b. melitensis. washing method did not differ in isolation rate with direct inoculation; however, brucella was isolated in a shorter period in washing method. conclusion higher isolation rate when compared with prior studies indicates an appropriate sampling time and technique, rapid inoculation to the media, and the lack of antibiotic therapy before sampling. washing method has the preference of shorter isolation time to direct inoculation; however, it is faced with a higher risk of contamination

Isolation, identification and antimicrobial susceptibility of brucella blood culture isolates.

UNLABELLED Isolation of slowly growing and fastidious Brucella spp strains from clinical specimens is difficult, because of varying factors, including species specificities, stadium of disease, and previous antibiotic treatment of the patients. The use of automated blood culture systems has overcome some cultivation problems. The automated identification system such as VITEK 2 compact allows mo...

Genome Sequences of Brucella melitensis, Isolated from Blood Samples of Brucellosis Patients in Malaysia

Human brucellosis is a neglected zoonotic disease and has widespread geographical distribution. Brucella melitensis has caused outbreaks and sporadic cases in Malaysia. Here, we present the whole-genome sequences of four B. melitensis strains isolated from brucellosis patients in Malaysia.

Immune reactivity of sera obtained from brucellosis patients and vaccinated-rabbits to a fusion protein from Brucella melitensis

Objective(s): Brucella spp. are facultative intracellular pathogens which can stay alive and multiply in professional and nonprofessional phagocytes. Immunity against Brucella melitensis involves antigen-specific CD4+ and CD8+ T-cells activation and humoral immune responses. Due to negative aspects of live attenuated vaccines, much attention has been focused on finding Brucella-protective antig...

Isolation of Brucella abortus and Brucella melitensis from Seronegative Cows is a Serious Impediment in Brucellosis Control

Brucellosis is a zoonosis occurring worldwide, with economic and public health impacts. Its diagnosis remains a challenge in endemic countries and basically relies on serology. The present study was carried out on two dairy cattle farms allegedly free from brucellosis, but with sporadic cases of abortion. The aim of this study was to investigate the presence of Brucella (B.) spp. in uterine dis...

تشخیص بروسلوز با استفاده از سیستم کشت خون ‌ BACTECو تائید نتایج آن با PCR

Abstract: Brucellosis is one of the most common zoonotic diseases in Iran. Growth of Brucella is slow and blood cultures of these bacteria are time-consuming via classical methods. We try to evaluate BACTEC 9120 system capacity in order to detect of bacteremia due to Brucella spp and to confirm isolated bacteria by PCR. Blood culture sample of 102 suspected patients evaluated by BACTEC 9120 sys...
